Profilaksis Ekstrak Brokoli terhadap Kadar Gula Darah pada Tikus putih yang Diinduksi Deksametason

Diabetes mellitus merupakan penyakit metabolik kronis dengan hiperglikemia kronis. Meningkatnya konsumsi deksametason, terutama saat pandemi COVID-19, menghasilkan peningkatan pasien hiperglikemia bahkan meningkatkan risiko terkena type 2 diabetes mellitus (T2DM). Glukokortikoid, termasuk deksametason, sering menyebabkan drug-induced hyperglycemia dan drug-induced diabetes. Terapi profilaksis dengan bahan alami, seperti brokoli, yang mengandung agen antidiabetes, antihiperglikemik, antioksidan dan antiinflamasi dari senyawa flavonoidnya diperlukan untuk mencegah atau menurunkan hiperglikemia dan risiko terjadi T2DM.Penelitian ini bertujuan untuk mengetahui pengaruh pemberian profilaksis ekstrak brokoli (Brassica oleracea var. italica) sebagai penghambatan terhadap peningkatan kadar glukosa darah puasa tikus yang disebabkan oleh induksi deksametason. Tikus putih (Rattus norvegicus) jantan galur Wistar sebanyak 24 ekor dibagi dalam 3 kelompok berbeda; K(-) = tanpa perlakuan, K(+) = induksi deksametason subkutan 10 mg/KgBB/hari selama 10 hari, KP = profilaksis ekstrak brokoli 1000 mg/KgBB/hari selama 7 hari dan ekstrak brokoli dilanjutkan bersamaan dengan induksi deksametason subkutan 10 mg/KgBB/hari selama 10 hari. Penelitian berlangsung selama 24 hari dan hari ke-25 sampel darah diambil untuk pengukuran glukosa darah puasa.
Didapatkan hasil rerata glukosa darah puasa K(-) = 173,62; K(+) = 304,89; KP = 180,50. Hasil uji parametrik One Way Anova didapatkan signifikansi p = 0,001(p < 0,05). Hasil Post Hoc LSD; K(+) dan KP memiliki signifikansi p = 0,001 (p < 0,05).
Pemberian profilaksis ekstrak brokoli 1000 mg/KgBB dapat menghambat peningkatan (menurunkan) kadar glukosa darah puasa tikus putih jantan galur Wistar yang diinduksi deksametason 10 mg/KgBB/hari selama 10 hari.
